Abstract

Bone and Osteoarthritis places emphasis on the molecular and cellular events that lead to osteoarthritis, stressing the role of subchondral bone, which distinguishes this from other books on the disease. A novel aspect is the attention given to the possible epigenetic basis, together with a discussion of the genetics predisposing to osteoarthritis. Detailed analyses are given of the role of the synovium, of the molecular mechanisms that lead to degradation of the cartilage matrix, of the hypertrophy of the cartilage cell, of the anabolic and catabolic roles of cytokines, may lead to novel approaches to clinical treatment, utilizing anabolic mediators or molecules that target steps in the disease process. Also discussed are animal models and how mechano-responsiveness is compromised by mechanical injury. Orthopedics and rheumatology have become close conceptually, as advances in bone and joint biology have enabled bench and translational scientists, as well as practitioners, to approach clinical problems comprehensively. Because bone plays a role in initiating osteoarthritis, therapeutic approaches focusing on bone tissue are included in the discussion of novel treatments. The general topic of osteoarthritis is therefore a timely subject for a series on bone biology. This book, intended for clinicians, researchers and students, provides information that will orient the novice and update the specialist. No other book treats the relationship of bone to osteoarthritis in similar fashion or provides a comparable underpinning of joint pathophysiology.

Abstract

Eukaryotic Membranes and Cytoskeleton: Origins and Evolution discusses the evolutionary origin and diversification of eukaryotic endomembranes and cytoskeleton from a cell biological and comparative genomic perspective. Many of the chapters present original research data from comparative genomic surveys. The presence/absence of gene families with central roles in endomembrane and cytoskeleton dynamics in a variety of eukaryotic taxa and an understanding of eukaryote phylogeny allow us to accurately reconstruct the cellular machineries present in the last common ancestor of eukaryotes. Such a reconstruction is fundamental if we are to understand eukaryotic diversification since this is the ancestral cell from which all diversity arose. Comparative genomics can likewise tell us which lineages expanded or reduced certain gene families and the associated cellular machineries.

Abstract

Our rapidly increasing knowledge about tumors of the brain and spine has rendered these tumors more treatable than ever before. In the new world of central nervous system oncology, maximally complete resection is followed by conformal radiotherapy and often by chemotherapy, the selection of which is based on the tumor’s susceptibility to drugs that have biological actions known to interact with the tumor’s particular molecular signature. Abstract

The most prominent naturalist in Britain before Charles Darwin, Richard Owen made empirical discoveries and offered theoretical innovations that were crucial to the proof of evolution. Among his many lasting contributions to science was the first clear definition of the term homology-"the same organ in different animals under every variety of form and function." He also graphically demonstrated that all vertebrate species were built on the same skeletal plan and devised the vertebrate archetype as a representation of the simplest common form of all vertebrates. Just as Darwin's ideas continue to propel the modern study of adaptation, so too will Owen's contributions fuel the new interest in homology, organic form, and evolutionary developmental biology. His theory of the archetype and his views on species origins were first offered to the general public in On the Nature of Limbs, published in 1849. It reemerges here in a facsimile edition with introductory essays by prominent historians, philosophers, and practitioners from the modern evo-devo community.
